You can use Pixel Check to look for bad pixels.

Other than that, simply click on the Apple icon at the upper left most corner and click on "about this mac", then "more info" to open the system profiler. Then check the specs are correct, RAM, HD size. Check that all the ports are working, LAN, Firewire, USB, modem. You don't need to do these things on the spot though. If ever anything is wrong, you have warranty so simply bring it back to them.

Most reseller are wired so ask them to download Pixel Check or similar apps. You would effectively check screen pixels and your ethernet connection by doing so. Bring blank CDs/DVDs to test your drive's read/write capabilities. Check USB/Firewire connections as well. If they have WiFi check that too or you can go to the nearest wireless site if you find it convenient. Finally if you have a bluetooth capable phone download Romeo and try it. This would save you a lot of trips back to the reseller.
Good Luck on your new purchase.
